L'Area Protetta comprende quattro distinti settori, tra Monterotondo, Mentana e Sant'Angelo Romano sui colli situati tra la valle del Tevere e i rilievi calcarei dei monti Lucretili. I primi due, nella parte occidentale, comprendono la vasta Macchia del Barco e sono separati tra loro da un'esile fascia non protetta dove transita il tracciato autostradale dell'A1 nel tratto della cosiddetta "bretella" Fiano-San Cesareo. Più ad est si trovano il Bosco Nardi, noto anche come Bosco di Grotte Cerqueta, appena a nord di Sant'Angelo Romano, nonché il piccolo settore che comprende la singolare voragine del Pozzo del Merro.
